Non-coding DNA for expression in S. cerevisiae to place between the upstream activating sequence and the core promoter.
Usage and Biology
This part was used in CCA_San_Diego’s 2021 CollaGene project to increase transcription of a bacterial collagen protein in yeast. It was added right after the upstream activating sequence (BBa_K3956000) and directly before a core promoter (BBa_K3956003 - consisting of a scanning region and a TSS) to increase expression based on the study design by Decoene et. al, 2019. Although we were able to verify that the construct had been correctly inserted into appropriate selective plates by colony PCR verification, the protein expression was not able to be verified within the time constraints of the competition.
We want to emphasize that this sequence may still be successful, but its efficiency was unable to be demonstrated within the time span that CCA_San_Diego had in the lab. For future teams, we advise testing construct designs with successful expression with and without the sequence to compare expression levels.
